Why do we typically use UV-vis for detecting proteins?
As three aa have double bonds, tryptophan, tyrosine and phenylalanine that absorb at certain light so if you know how many of that aa you can know how much protein there is.

What is the beer-lambert law and what is it used for?
absorbance=E x l x c . L-->length light passes through solution cm. C--> conc in molar. The law shows the absorbance of light and the conc of the solution absorbing it.

Describe IR spec.
uses longer wavelengths and lower freq than uv-vis. it is associated with bond vibration, stretching and bending. Limited to macromolecules due to low resolution.

Describe fluorescence spec.
excite samples it absorbes light at certain wavelength and gives light out at anout (detector at 90degrees). Very sensitive can detect a single molecule. fluorophores typically contain multiple aromatic groups.

Describe NMR spec
very insensitive. The nuclear spin of an atom shows the chemical environments as it effects freq of nuclei. atomic resolution. individual peaks are one atom. so high resolution power

measure mass of a molecule to identify a sample

what is x-ray crystallography
pattern of crystalline solid exposed to x-ray radiation to identify it's 3D structure.
