Measurement of drug concentration

?
What different methods of measuring drug concentration?
Physicochemical - mass spec, IR/UV spectroscopy. Bioassay-2x2 bioassay. Immunoassays-interaction of antigen/ab. radioimmunoassay, enzyme-linked immunoabsorbent assay
1 of 5
Describe Bioassay.
measuring drug conc by measuring response in relation to a standard preparation. log M=EC50 test/EC50 standard. if M was 10 you would need 10 more test drug, so much have 10x less active substance.
2 of 5
Describe an enzyme-linked immunoabsorbent assay ELISA-antigen detection
To detect antigen-ab coated filter, wash, add soltuion, wash, add second ab with enzyme that is against antigen. incubuate with enzyme substrate. if colour forms then hormone present
3 of 5
ELISA-non competitive to detect antibody
antigen coated, incubate with sample, wash, incubate with enzyme linked to 2ab (against 1 ab) if colour then antibody present. Same for competitive ELISA but if colour produce no antibody present.
4 of 5
Describe a radioimmunoassay.
The more hormone that binds the less radioactivity is detected. Use standard curves.. Incubate with unknown hormone, incubate with radioactive hormone, measure radioactivity. hormone will push radiolabelled hormone out.
