Tamoxifen- mechanism of action
Estrogen- receptor nuclear complex → recruits dna repressor protiens such as NCor1 and NCoR2 → these bind to the DNA and inhibit HER2 expression → cell growth inhibited
1 of 12
Tamoxifen- therapeutic affect
repression of the HER2 pathway will result in reduced proliferation of cells tumour shrinkage, prevent increase in size, reduce proliferation and differentiation
2 of 12
Herceptin (monoclonal antibody)- mechanism of action
Binds to HER2 and prevents its dimerization causes optimization marking the cells --> allows for the immune system to attack and kill → activate the macrophages, ADCC pathway and complement cascade
3 of 12
Herceptin (monoclonal antibody)- therapeutic affect
destruction by immune cells and immune mediators Prevent growth and proliferation of tumour cells
4 of 12
Diamorphine- therapeutic affect
result in increase of enkephalins and enorphine to desensitize nococeptors- reduce pain
5 of 12
Diamorphine-mechanism of action
mu-opioid agonist- active 6-monoacetylmorphine (6-MAM), and then to morphine, which bind to μ-opioid receptors
6 of 12
Haloperidol- mechanism of action
Selectively bind the D2 receptors in mesolimbic and mesocortical area→ antagonizes the dopamine pathways by inhibiting receptor binding (+) binds to 5-HT2 receptors and prevents serotonin binding in the hippocampus and pre-frontal cortex (-)
7 of 12
Haloperidol- therapeutic affect
manage the positive and negative symptoms by anatgonising dopamine in the mesolimbic and mesocortical and serotonin in hippocampus and pre-frontal cortex
8 of 12
Diazepam- mechanism of action (anticonvulsant)
Anticonvulsant → binds to the voltage-dependent sodium channels and prevents their opening so inhibits sustained firing
9 of 12
Diazepam- therapeutic affect
anti anxiety, anti-convulsive, muscle relaxant
10 of 12
Diazepam- mechanism of action (muscle relaxant)
Muscle relaxant → Inhibition of the polysynaptic pathways in the spinal cord therefore sensory motor systems are disrupted
11 of 12
Diazepam- mechanism of action (anti-anxiety)
bind nonspecifically to benzodiazepine receptors. coupled to gamma-amcoupled to(GABAA) receptors, this increases GABA affinity. Binding of GABA opens the cl- channel--> hyper polarisation and prevention of further excitation
