Topic L microbiology and human impact on the environment

?
How are bacteria classified?
Classified according to their Shape, cell wall structure and metabolic, genic and antigenic features
1 of 10
What are the names of some shaped bacteria?
spherical (coccus) rod shaped (bacillus) spiral (spirillum)
2 of 10
what do the names of these bacteria indicate?
Can indicate the disease they cause
3 of 10
What does the gram staining technique show?
Shows cell wall structure of bacteria and determines if its gram + or gram -
4 of 10
What does the antibiotic penicillin prevent in terms of bacteria?
Prevents cross links from forming within the peptidoglycan layer weakening the cell wall`
5 of 10
What type of bacteria is most affected by penicllin?
Gram + bacteria becomes subject to osmoticlysis when water enters the bacterial cell causing the cell to burst
6 of 10
Features of gram + bacteria
Thicker cell wall
7 of 10
Gram + bacteria properties
-Thicker cell wall -Thick layer of peptidoglycan -No Lipopolysaccharide layer vulnerable to penicillin and lysozyme action -Peptidoglycan layer retains crystal violet stain so stains purple -Staphylococcus and streptococcus
8 of 10
Gram - bacteria properites
-Thinner cell wall -Thin layer of peptidoglycan -LPS layer protects against penicillin and lysozyme action -LPS layer prevents uptake of crystal violet stain, only stains red once LPS removed and counter stained -Salmonella E.coli
9 of 10
Descirbe the gram stain procedure
-Transfer sample of bacteria to a glass microscope slide using an inoculating loop Pass slide through bunsen burner flame to fix bacteria to slide also kills them - Add few drops of crystal violet stain and leave for 30 secs - Rinse using water
