D.R = apply x5 lesions, 5-10 days, 1cm to eyes. 400mg up to x3 day

C = renal failure, hypersensitivity, immunocompromised patients

S.E = Nausea, Vomiting, Diarrhoea, eye pain, eye inflammation
T = coma, seizures, lethargy

Mechanism of Action?
M.o.A = conversion to aciclovir triphosphate, inhibiting viral DNA polymerase causing DNA chain termination

Absorption? Distribution?
A = 10-20% oral bioavailability, decreases with increased doses. <0.02-9.4% topical. buccal + opthalmic minimal. food doesn't affect bioavailability
D = 0.6L/kg. 9-33% plasma protein bound

Metabolism? Elimination?
M = <15% oxidised to 9-carboxymethoxymethylguanine. 1% 8-hydroxylated to 8-hydroxy-aciclovir
E = 90-92% via glomerular filtration + tubular secretion in urine + unchanged. <2% faeces. <0.1% CO2 expiration
