What is a neuromuscular junction?
The junction between motor nerves and skeletal muscle. one motor nerve controls multiple skeletal muscle fibres. The nerve sits in junctional cleft. The membrane in folds to increase SA for receptors, schwann cells lie across top of cleft

How is depression treated?
Selective serotonin reuptake inhibitors SSRIs prevent uptake of serotonin to treat depression.

Describe the synthesis of acetylcholine.
CAT enzyme transfers acetyl coenzyme A and adds it to chloine. ester bond formed. chloine taken up by transporter molecule from outside cell, ACh into vesicles. Ca2+ causes exocytosis

What prevents ACh ?
Hemicholiniums inhibit transporter protein that takes up choline. It takes a while to have effect as ACh already stored. Vesamicol-interferes with process of putting ACH into vesicles. Botox-blocks transmission at terminals, as inhibits exocytosis.

Pancuronium-reversible competitive antagonist. prevents ACh binding. Suxamethonium-initially stimulates but prolongs depolarisation so another ap can't be fired. muscles relax.

How does ACh activate a receptor?
2 molecules has to bind to each alpha subunit, brings about a conformational change, channel opened a non-selective cation channel and membrane becomes depolarised. Activates voltage-gated Na channel to form an ap.
