LD50/ED50 or TD50/ED50 The larger the number the greater the safety of the drug., however doesn;t show overlap. eg high efficacy dose may mean small toxicity.

metabolised-combination with sulfate and glucuronic acid/oxidation and glutathionine.

Overdose of paracetamol
depletion of glutathionine, reacts with liver proteins hepatic necrosis and death

Metabolism of aspirin
metabolised by esterases to salicylic acid then conjugated with glucuronic acid/glycine. Salicylate interferes with function of ETC in mitochondria.-hyperventilation etc- metabolic acidosis can be fatal
