What does agar jelly contain?
Carbohydrates for energy, amino acids and nutrients for growth

How do you sterilise agar?
In a bottle, in a pressure cooker at above boiling

How do you sterilise an inoculating loop?
In hot flames until it glows red

Once you have inoculated the agar jelly, what do you do?
Seal the Petri dish with Sellotape to avoid contamination from the air and incubate in an oven at 25C to help the bacteria reproduce

Why do you put the inoculated agar jelly in the fridge after 24 hours?
To limit the numbers of bacteria

The agar has a limited supply of food

How can you reduce contamination from the air?
Only open the Petri dish slightly when inoculating, do not breathe on the agar, Sellotape the dish shut afterwards
