How do cells become specialised?

To become specialised cells differentiate. Differentiation is where a cell changes to become specialised for the job. As the cells change, the subcellular structures change and become different types of cells, this then allows them to carry out specific functions. 

Most of the time, this happens as an organism develops, before they specialise as is the case with most animal cells. However, plant cells keep this ability for their whole lives.

In humans, the cells that differntiate in adults are skin and blood
