I = pain + inflammation, rheumatic disease, dental pain, migraines, mild - moderate pain (dysmenorrhoea)
C = active GI bleeding, varicella infection, heart failure

S.E = nausea, headaches, GI discomfort, hypersensitivity, skin reactions
T = epigastric pain, tinnitus, vomiting

Ibuprofen:
Mechanism of Action?
M.o.A = inhibits prostaglandin (pain/fever) + thromboxane (blood clotting stimulators) synthesis via arachidonic acid pathway

Ibuprofen:
Absorption? Distribution?
A = rapidly in upper GI tract (oral), peak conc. 1-2 hours post extravascular administration. Rate decreases with food consumption
D = vol. 0.1L/kg. 99% plasma protein bound (saturable + non linear), conc. greater 20mcg/ml

Ibuprofen:
Metabolism? Elimination?
M = via liver through hydroxylation + carboxylation, metabolites in cp450 system produces 1-hydroxyibuprofen. Conjugation -> glucuronide (oxidative metabolites)
E = 90% via urine. biliary excretion - 1% unchanged. 99% after 24 hours
