What is the res of a light microscope?
200nm
5 of 15
How do you calculate the image size of a sample?
magnification x actual size
6 of 15
What microscope is used to view in 3D?
An SEM
7 of 15
What is an artefact?
A false image produced when prepping a slide
8 of 15
Why is staining used in microscopy?
To help increase contrast of the structures viewed
9 of 15
How would you prepare a wet mount?
Place the droplet int he centre of the slide. Then place the cover slip at an angle on the droplet to reduce the chance of bubbled forming under the cover slip
10 of 15
How do you prepare a dry mount?
Section the specimen if needed or keep it whole. Then place it in the centre of the slide and cover it with a cover slip.
11 of 15
What are squash slides used for?
Soft samples. Root tip squash slides are used for investigating cell division.
12 of 15
How do you prepare a sample for staining?
Placed on the slide and air-dried. It is then passed through a flame to heat fix it onto the slide and will then take up stains
13 of 15
What is the Gram stain technique used for?
Separating bacteria into two groups:Gram-positive and Gram-neg bacteria by using crystal violet, iodine and alcohol
14 of 15
What is the Acid-fast technique used for?
To differentiate between species of Mycobacterium from other bacteria by using a lipid solvent and acid
