Set up five water baths at temperature of 10,20,30,40 and 60

Step 2:
Add 2cm3 of lipase into a test tube, label the test tube with a temperature and place them in the corresponding water bath

Step 3:
Label a boiling tube with the temperature you will be investigating

Step 4:
Add 5 drops of phenolphthalein into a boiling tube

Step 5:
Measure out 5cm3 of milk using a measuring cylinder (or syringe) and add this to the same boiling tube

Step 6:
Measure out 8cm3 of sodium carbonate solution using another measuring cylinder (or syringe) and add this to the boiling tube. To the same test tube add 3cm3 of pH 7 buffer. The solution should now be pink

Step 7:
Place a thermometer in the boiling tube. Take care as the equipment could topple over

Step 8:
Place the test tube in a water bath and leave until the contents reach the same temperature as the water bath

Step 9:
Remove the thermometer from the test tube and replace it with a glass rod

Step 10:
Measure out 5cm3 of the solution from the boiling tube and add this to the lipase solution

Step 11:
Stir the contents of the test tube until the solution loses its pink colour

Step 12:
Stop the clock/watch and note the time in a suitable table of results

Step 13:
Plot a graph of time taken for the reaction to occur against temperature

Step 14:
You can convert this to a rate of reaction graph by calculating 1 divided by time for each of the temperatures. (If any tubes have not reacted in the time taken, this is a rate of zero.)
