What is the process of Direct ELISA?
An antibody attached to an enzyme is added to an antigen. This antigen-antibody complex is put through a wash and a substrate is added which forms an enzyme-substrate complex with the enzyme on the antibody.

What will a colour change show?
Antigen/Antibody is present within the sample that was tested.

What does the intensity of the colour change represent?
quantity of antigen/antibody present

What is the process of indirect ELISA?
Sample containing different antibodies is added to a well containing a fixed antigen. Specific antibodies will bind to antigen.Well is then washed.Secondary antibody is added and binds to primary. Well is washed.Substrate is added in solution.

Why is the well washed?
To remove unwanted antibodies from the well

What shows a positive result for indirect ELISA?
Enzyme on secondary antibody will bind to substrate in solution to produce enzyme substrate complex and there will be a colour change.

Which process is shorter and why?
Direct ELISA is quickest as the process is shorter

Which form of ELISA is more definitive and why?
Indirect ELISA is more accurate and definitive because an antigen is more likely to bind to antibody when it is fixed rather than the antibody.
