What 4 requirements are needed for semi conservative replication to take place?
4 types of nucleotides each with their bases must be present, both strands of DNA molecule act as a template for the attachment of nucleotides, the enzyme DNA polymerase and a source of chemical energy is required.

What are the steps of semi conservative replication?
DNA helicase breaks down the H2 bonds which link the base pairs of DNA. This then causes the double helix to separate into 2 strands which both acts as a template. Free nucleotides attach to base pairing. DNA polymerase joins the nucleotides.

The DNA polymerase joins the nucleotides together in a condensation reaction and this is when the H2 bonds reform. The new DNA molecules contain one old strand and one new strand which is called semi conservative method.

Why is the replication of DNA described as being semi conservative?
As there is one new strand but one old strand is kept as each strand acts as a template.

Why can new nucleotides only be added in a 5 to 3 direction?
Because the DNA polymerase joins the nucleotides which are specific and are only complimentary to the 5 end of the strand. The shape of the 5 end and the 3 end are different so the new nucleotides cant bind onto the 3 end first as not complimentary.
