Practical biochemistry tests

?
what is the test for starch?
add iodine reagent. positive result is blu-black
1 of 18
what is the test for protein?
add biuret solution. positive result is lilac
2 of 18
what is the test for fats
add ethanol. positive result is cloudy emulsion
3 of 18
what is the reagent for test for reducing sugar?
benedicts solution
4 of 18
what is the procedure for the test for reducing sugars?
add benedicts and then boil at more than 80c. positive test is green, yellow or brick red
5 of 18
what is the reagent for the test for non-reducing sugars?
add hydrochloric acid then sodium hydroxide and finally, benedicts solution
6 of 18
what is the procedure for the test for non-reducing sugars?
do the test for reducing sugars first. add Hcl to break glycosidic bonds then neutralise with sodium hydroxide then boil with benedicts
7 of 18
what is the result for the test for non-reducing sugars?
no change at first but after adding Hcl there will be a normal colour change
8 of 18
what is colorimetry used for?
when quantifying the concentration of sugar in the original solution when there are changes
9 of 18
what are the stages of colorimeter?
fill in later
10 of 18
what are the four stages of creating a calibration curve?
1. take a series of known concentrations of reducing sugar 2. carry out benedicts test 3. use a colorimeter to record percentage transmission of light through the supernatant 4. plot a graph of transmission against concentration of sugar
11 of 18
How do biosensors work?
they take a variable that cannot be easily measured and convert it to an electrical signal
12 of 18
What four things can biosensors be used for?
detect contaminants in water, pathogens, toxins in food and airborne bacteria.
13 of 18
what are the two key components in chromatography?
stationary phase and the mobile phase
14 of 18
what is the stationary phase?
this is the chromatography paper made of cellulose or the TLC plate made of plastic with a later of silica gel
15 of 18
what is the mobile phase?
this is the solvent for the biological molecules
16 of 18
what is the Rf value?
distance travelled by solute / distance travelled by solvent
17 of 18
what three solutions can you use for chromatography if solutions are colourless?
UV light for TLC chromatography, Ninhydrin for amino acids and Iodine
