Observing unstained specimens
Some microscopes use light interference (rather than light absorption) while others use a dark background against which the illuminated specimen shows up.

Coloured chemicals that bind to molecules in or on the specimen, making it easier to see.

Types of stains
Some stains are all-purpose, for example methylene blue; however, some are differential stains, which bind to specific cell structures.

Differential staining
Acetic orcein binds to DNA and stains chromosomes dark red. Eosin stains cytoplasm. Sudan red stains lipids. Iodine stains cellulose yellow and starch granules blue/black.

Observing prepared specimens
Preparing slides includes dehydrating the specimen, embedding it in wax to prevent distortion during slicing, using a special instrument to make very thin sections, then staining and mounting the specimen in a preserving chemical.

Calculations involving magnification
Magnification = Image size / Actual size

Drawing slides
When drawing slides use a sharp pencil, indicate the scale, and make a lower plan showing where different tissue areas are (not individual cells); use clear, unbroken lines and do not shade. Label with straight, horizontal lines to the structure.
