In what type of organism would you find rubredoxin?
Bacterial
1 of 15
What is the function of rubredoxin?
Electron transfer/ conversion of SO to H2O2
2 of 15
What does the active site of rubredoxin look like?
FeII/FeII 4x S-Cys
3 of 15
What type of donors do rubredoxin have?
thiolatr donors -SCH2
4 of 15
How is the strain achieved in rubredoxin?
B-sheets cause cysteine residues to be rigid, causing whole enzyme to be rigid.
5 of 15
How do we know whether the Entatic effect has a role in rubredoxin?
The mononuclear version of the active site has longer bond lengths. The metal is distorted in the enzyme so that the bond lengths are between that of FeII and FeIII.
6 of 15
What does the distorted bond lengths in rubredoxin cause?
rapid cycling between oxidation states.
7 of 15
What are the spin states of the metal in Rb?
Fe(II)= high spin , Fe(III)= high spin
8 of 15
What are the colours of the oxidation states in Rb?
Fe (II) RED, Fe(III) colourless
9 of 15
What is the redox potential of Rb and why?
-60mV, hard to reduce due to negative charge caused by thiolate donors.
10 of 15
What is the structure of ferritin?
24 protein subunits, globular protein, alpha helices form hollow cages, each sub unit has a di-iron active site with ligands from each of the 4 alpha helices.
11 of 15
What is ferritin?
A super family of protein caged Fe2o3 h20 biominerals
12 of 15
What is the uptake mechanism of ferritin?
1) Transferrin has transported Fe2+ in to the cytoplasm 2) Ferritin catalyses the oxidation of Fe(II) to Fe(III). 3) This is done with oxygen and forms Fe(III)O minerals 4) When the concentration of the Fe(III) oxides increases crystallisation
13 of 15
Why is ferritin such a complex structure?
Ferritin is complex so that the reaction of Fe and O can be controlled as Fe & O would crystallise straight away upon contact.
14 of 15
What is the function of ferritin?
1) Oxidise Fe(II) to Fe(III) and form minerals. 2) Mineral dissolution and exit of Fe(II) when required.
