Ag-Ab binding forces?
Ionic Bonding, Hydrogen Bonding,Hydrophobic Bonding, Van der Waal forces
1 of 14
Affinity constant and unit
K=Ag-Ab/ (Ag)*(Ab), delta G=-RTlogek
2 of 14
Avidity definition
Overall Binding affinity (more than one binding on an antigen)
3 of 14
What is Bonus Effect
loosen of one binding can be recovered more easily if the Ab and Ag are still bound to each other by other binding
4 of 14
Cross Reactivity Definition
Binding of Ab to a protein which was not originally used as the antigen to produce Ab (e.g. Human anti-insulin and Pig anti-insulin). Partial compatible within the molecule and epitope.
5 of 14
Use of cross reactivity
The means used to compare the structure of two proteins
6 of 14
What are the application of Ag-Ab interaction
Immunologic assays and production of Ab
7 of 14
What is an assay
A lab method to detect the composition, quantity or quality of one more components
8 of 14
Use of immunoassays
1. To detect Ab level in blood of patients suffering from infection 2. To detect antigens such as virus or hormone in a biological sample. 3. Characterization of Ag and Ab
9 of 14
Principle of immunoassays
Ab-Ag interaction. Primary ag-ab interaction is on molecular level so not visible. Secondary interaction is visible.
10 of 14
What are secondary Ab-Ag interaction (4 immunoassays)?
Precipitation, agglutination, complement fixation, or the use of labeled reagents.
11 of 14
Precipitation principle and conditions
1. Ag and ab conc. need to be equivalent. Too high in either conc will result in no result due to postzone or prozone effect.
12 of 14
Agglutination principle
make use of the fact tat Ag are naturally occurred on cell surface
13 of 14
Complement fixation principle
Make use of the animal defense mechanism carried by complement proteins which binds onto any ag-ab complex on cell surface and forming a pore to lyse the cell.
