Polymerase Chain Reaction:

Equipment:

  • DNA fragment to be copied.
  • DNA polymerase, capable of joining nucleotides together. This enzyme has been taken from bacteria in hot springs so it is thermostable.
  • A primer, which is complimentary to the bases at the two ends of the DNA.
  • Free nucleotides.
  • Thermocycler, which is a temperature regulating computer.

Step 1) All equipment is placed in a vessel in the thermocycler at 95oC. This breaks the DNA fragment up into single strands.
