Polymerase Chain Reaction:
Equipment:
- DNA fragment to be copied.
- DNA polymerase, capable of joining nucleotides together. This enzyme has been taken from bacteria in hot springs so it is thermostable.
- A primer, which is complimentary to the bases at the two ends of the DNA.
- Free nucleotides.
- Thermocycler, which is a temperature regulating computer.
Step 1) All equipment is placed in a vessel in the thermocycler at 95oC. This breaks the DNA fragment up into single strands.
Step 2) The system is cooled to 55oC in order to allow the primers to anneal/attach to the DNA. This then allows for DNA polymerase…
