• the tonoplaast cell surface membrane needs to be destroyed
• partially permeable membrane

Limitations and Effect 

• replicates -- not possible to see if results are reliable
• Discs not left long enough -- underestimate of pigment released
• same beetroot of region of beetroot not used --different material contains diferent amount of pigment to release
• Difficult to match…
