Virulence Determinants

Enxymes: Cause damage to host, for example; gas gangrene, has necrotising fascitis.

Toxins: Exotoxins is secreted by bacteria and each has a specific effect. Endotoxins are part of the cell wall and are only released when the cell dies. Lots of bateria produce endotoxin and all have the same effect- fever, shock, rash and even death e.g. meinigitis where extra endotoxin is made and released into the body.

Survival inside cells: This stops our defence from attacking the invader. Tuberculosis bacteria do this, as they have a waxy cell wall which helps protect them.

Medical Microbiology

Micro-organisms are ubiquitous (everywhere) so interact with the immediate environment. They get their nutrients from the environment then release waste into the environment. This can be beneficial e.g. Yeast, pharmaceutical uses.

Normafluora-organisms that are present but depending on the environment causes disease that can sometimes benefit you.

Oportunistic Pathogen- causes infection under certain conditions.

Putting the theory to the test:

4. Organism should be re-isolated and shown to be the same as the original.

Biofilms

Biofilms are assemblages of bacteria cells adhered to a surface and unclosed in an adhesive matrix excreted by the cells. The matrix consists of a mixture of polysaccharides (chains of carbohydrates)

Biofiulms trap nutrients from microbial growth and help prevent detachment of cells in flowing systems.

Example;
Pseudomonas aeruginosa

  • Produces biofilms
  • Intracellular communication (quorum sensing) is critical in the maintenance of a biofilm
  • Interspecies signalling (communication within species) and Intraspecies signalling
  • increased resistance to antibiotics

Limitations

Biofilms have been implicated in several medical and dental conditions e.g. periodontal disease(affects soft and hard structures that support the teeth), kidney stones and others.

In industrial settings biofilms can slow the flow of liquids through pipelines and can accelerate corrosion of inert surfaces.

Filamentous fungi- generally appearing as a circular colony that may be cottony or wooly but with filaments that are not organized into large fruiting bodies, such as mushrooms.

Lag phase-transition phase for organisms ot ajust to the new environment. length of lag phase depends on the previous environment e.g. pH, growth medium, temperature etc. to reduce the time in lag phase create optimum conditions to increase productivity.

Exponential Phase (Log phase)- period of balanced growth, biomass increases logarithmically. Growth is characterised by increase in cell number and increase in cell size. maximum growth rate is determined by conditions e.g. carbon and energy sources, temperature, pH, water availability etc. 

Deceleration Phase- rate of growth decreases due to nutrient limitation (e.g. C,N or P source) and toxicity/stress e.g. too many organisms, oxygen depletion, over production of acid/alcohol.

Stationary phase- is the decrease in primary metabolic activity, production of secondary metabolites e.g. antibiotics, decrease in cell viability, cell walls start to leak leading to death.

Psychrophile-
extremophilic bacteria or archaea which are cold‐loving, having an optimal temperature for growth at about 15°C or lower, a maximal temperature for growth at about 20°C and a minimal temperature for growth at 0°C or lower

Mesophile- an organism that can grow best in moderate temperatures typically between 20-45°C (Humans)

Thermophile-a bacterium or other microorganism that grows best at higher than normal temperatures.

This can occur due to lipid content of membrane and enzymes.

  • Prokaryotic microorganisms- no organelles, reproduce asexually, nucleus isn't covered with a membrane.

They consist of either DNA or RNA surrounded by a protective coat of protein to create genetic material.

Bacteria are unicellular prokaryotic microorganisms 

They can; 

  • Independently metabolise

1. Metabolism
Uptake nutrients from the environment, their transformation within the cell, and elimination of wastes into the environment. The cell is thus an open system.

Metabolic activities make cells chemically very complex and diverse. 

2.Reproduction (growth)
Chemicals from the environment are turned into new cells under the direction of pre-existing cells. 

Reproduction is based on:

a) replication of DNA and 
b) equal distribution of replicated DNA into two cells after division of the initial cell. 

Reproduction is the fundamental feature of the life. Ensuring existence of species and their evolution. 

3) Differentiation 
Formation of a new cell structure such as a spore, usually as part of a cellular life cycle. Differentiation is not a compulsory property of all microbial cells,but those species that form spores survive better in unfavourable conditions. 

4) Communicaiton 
Cells communicate or interact primarily by means of chemicals that are released or taken up. 
One of these consequences of cells communications is the quorum sensing mechanism. 

5) Movement 
Living organisms are often capable of self-propulsion. 
Motility is not compulsory propert of all microbes 

6) Evolution 
Cells contain genes and evolve to display new biological properties. Phylogenetic trees show the evolutionary relationship between cells.
