Root Tip Squash
1. Cut final 5mm off root tip from a plant such as an onion
2. Add stain e.g. orcein/toluidine blue and usually acid
3. Gently macerate (break up) on a microscope slide and squash carefully
4. Add more stain and warm then look at slide through microscope
Where mitosis occurs in root
Stain highlights chromosomes + acid softens tissue
Spreads out cells so layer 1 cell thick ­ easier viewing
Warmth intensifies stain
